Routine screening of fixed nematodes mounted in slides
with coverslip

Routine screening of heat-killed nematodes in cavity slides
without coverslip
Specimens are killed at 65C on a cavity slide, and captured using
Configuration
2 with its 100x/NA 1.0 water immersion objective and 1x/1.25x/1.6x/2x
magnification changer. The resulting clips at 100x, while definitely inferior
to results that would be obtained with oil immersion objectives, are nevertheless
good enough to allow routine screening, identification and VCE archiving
of nematodes straight from extract, without any fixation or mounting under
coverslip.
We do not provide an image map for these specimens, as this step would
obviously be too time-consuming in a situation where tens or hundreds of
nematodes were processed in a routine sample screening.
