Structure and Function in Insects

 

Recipes

2.5% Cobalt Solution

-0.25 g CoCl2

-10 ml of distilled water

It may be stored for a long time

Ammonium Sulfide solution

-5 ml Ringer

-5 drops of ammonium sulfide, concentrated ((NH4)2S)

Prepare when needed only.

Holder AAF

-80 ml 10% ethanol                                                       -8 ml glacial acetic acid

-12 ml 40% formaldehyde                                             It may be stored for a long time

Revealing base solution

-100 ml distilled water                                                 -3 g Arabic glue  

-10 g glucose                                                              -Shake well.  Take to PH 2.5 - 2.6 with citric acid. It can

-0.17 g hydroquinone                                                  be stored for a few days in a fridge.

-1.4 g citric acid

Revealing solution (with silver)

-10 ml revealing base solution

-1 ml AgNO3 1%

Prepare only when needed

Ringer solution

Base solution:

-1000 ml distilled water                                   -0.35 g KCl

-7.5 g NaCl                                                    -0.21 g CaCl2

-0.14 g NaHCO3                                           -2.5 g glucose

take to PH 7.0

Buffer:

-A: solution 0.15 molar KH2PO4

-B: solution 0.15 molar Na2HPO4

For the buffer, mix 19 volumes of A + 31 volumes of B.

Ringer: 10 volumes of base solution + 1 volume of buffer
