Structure and Function in Insects

 

Recipes

Special Buffer:

-Solution A of buffer: Take 11.30 g of (NaH2PO4H2O) to 500 ml with distilled water.  Dissolve.

-Solution B of buffer: Take 2.52 g of Sodium Hydroxide (NaOH) to 100 ml with distilled water. Dissolve.

To prepare the buffer, mix 83 ml of Solution A with 17 ml of Solution B.  Take to pH=7.30 with CIH or NaOH.  For each 100 ml of buffer, add 2.2 g of d-glucose and 0.9 ml of Cl2Ca 1% solution to adjust the osmotic pressure.

Special Fixer:

-Glutaraldehyde 2.5% and Paraformaldehyde 2.0% in special phosphate buffer.

-To prepare the fixer, dissolve 2g of Paraformaldehyde in 17 ml of Solution B of buffer in an Erlenmeyer, in some hot water that must not exceed 60 C.  when it is completely dissolved, add 83 ml of Solution a of buffer.  To complete, add 10 ml of Glutaraldehyde 2.5%.  Finally, adjust to pH=7.30.  For each 100 ml of fixer, add an additional 2.2 g of d-glucose and 0.9 of Cl2Ca 1% solution to adjust the osmotic pressure.

Hard Durcupan:

-Component A, 11.3 g

-Component B, 10 g

-Component C, 0.2 g

-Component D, 0.5 g

-Mix component A with B in magnetic mixer.  Add component D and mix for 30 minutes.  Add component C and mix for one hour.  Save in portions of 2-5 ml in a freezer at -20 C.

Blue metilene solution:

-Distilled water, 50 ml

-Sodium Borate (Borax), 0.5 g

-Blue metilene, 0.5 g

Mix and filter.
